1.9 Kb Aerolysin [hemolysin] encoding DNA was cloned from clinical isolate of Aeromonas hydrophila. The Aerolysin gene product was extracted from the recombinant E. coli clone and from A. hydrophila.
purification was accomplished by acid precipitation and ion -exchange chromatography on QAE- sephadex A- 50 followed by gel filtration on sephadex G- 100. The purified native enzyme had a specific activity of 132129 u/mg protein while for the recombinant one it was 363 I2 dmg. On both cases single band was observed on polyacrylamide gel electrophoresis with an estimated molecular weight of 60 kd or 56 kd respectively. The optimum temperature of the enzymatic activity was 37degree C. Papain was found to inhibit such activity
